Phenotypic divergence of Homo sapiens is driven by the evolution of human-specific genomic regulatory networks via two mechanistically distinct pathways of creation of divergent regulatory DNA sequences

چکیده
Running title: Divergence pathways of human-specific regulatory sequences Abstract Thousands of candidate human-specific regulatory sequences (HSRS) have been identified, supporting the hypothesis that unique to human phenotypes result from human-specific alterations of genomic regulatory networks. Here, conservation patterns analysis of 18,364 candidate HSRS was carried out based on definition of the sequence conservation threshold as the minimum ratio of bases that must remap of 1.00. A total of 5,535 candidate HSRS were identified that are: i) highly conserved in Great Apes; ii) evolved by the exaptation of highly conserved ancestral DNA; iii) defined by either the acceleration of mutation rates on the human lineage or the functional divergence from nonhuman primates. The exaptation of highly conserved ancestral DNA pathway seems mechanistically distinct from the evolution of regulatory DNA segments driven by the species-specific expansion of transposable elements. Present analysis supports the idea that phenotypic divergence of Homo sapiens is driven by the evolution of human-specific genomic regulatory networks via two mechanistically distinct pathways of creation of divergent sequences of regulatory DNA: i) exaptation of the highly conserved ancestral regulatory DNA segments; ii) human-specific insertions of transposable elements.

Mechanistically Distinct Pathways of Divergent Regulatory DNA Creation Contribute to Evolution of Human-Specific Genomic Regulatory Networks Driving Phenotypic Divergence of Homo sapiens
Thousands of candidate human-specific regulatory sequences (HSRS) have been identified, supporting the hypothesis that unique to human phenotypes result from human-specific alterations of genomic regulatory networks. Collectively, a compendium of multiple diverse families of HSRS that are functionally and structurally divergent from Great Apes could be defined as the backbone of human-specific ...
متن کاملCloning and sequencing of rainbow trout (Oncorhynchus mykiss) interferon regulatory factor 7
Interferon regulatory factor 7 (IRF7) gene was cloned from a subtractive cDNA library constructed with mRNAs obtained from rainbow trout (Oncorhynchus mykiss) macrophage cell line (RTS-11). Using expressed sequence tag clones of submitted IRF7 amino acid sequences, specific primers were designed. Results showed that IRF7 cDNA contains an ORF of 1251 nucleotides that translates into a 416 resi...

Changes in gene expression and regulation--due in particular to the evolution of cis-regulatory DNA sequences--may underlie many evolutionary changes in phenotypes, yet little is known about the distribution of such variation in populations. We present in this study the first survey of experimentally validated functional cis-regulatory polymorphism. These data are derived from more than 140 pol...
متن کاملAdaptive sequence evolution is driven by biotic stress in a pair of orchid species (Dactylorhiza) with distinct ecological optima
The orchid family is the largest in the angiosperms, but little is known about the molecular basis of the significant variation they exhibit. We investigate here the transcriptomic divergence between two European terrestrial orchids, Dactylorhiza incarnata and Dactylorhiza fuchsii, and integrate these results in the context of their distinct ecologies that we also document. Clear signals of lin...
متن کاملRapid sequence evolution of transcription factors controlling neuron differentiation in Caenorhabditis.
Whether phenotypic evolution proceeds predominantly through changes in regulatory sequences is a controversial issue in evolutionary genetics. Ample evidence indicates that the evolution of gene regulatory networks via changes in cis-regulatory sequences is an important determinant of phenotypic diversity. However, recent experimental work suggests that the role of transcription factor (TF) div...
